Design: Samples of 50 adults were homogenized in liquid nitrogen and RNA was extracted using the RNEasy Mini (Qiagen) kit. cDNA libraries were then prepared using 250–500 ng of total RNA. Briefly, mRNA were purified using oligo-dT primer coated magnetic beads (Invitrogen). Double stranded cDNA was synthesized using SuperscriptII reverse transcriptase (Invitrogen) and random hexamer primer (Invitrogen) and purified using the Ampure XP kit (Invitrogen). Illumina sequencing adapters were then ligated to the cDNA. The resulting PCR product was then purified with magnetic beads (Invitrogen), electrophoresed through a 2% agarose gel following excision of 300–500 nucleotide PCR products. The libraries were sequenced with paired-end protocol (2 ? 101 bp) using Illumina Hiseq2000 and TruSeq SBS chemistry. Demultiplexing was performed using CASAVA-1.8.2 (Illumina).

Opisthorchis felineus (Rivolta, 1884) or European liver fluke is epidemiologically important parasite that infect mammals including humans. O. felineus is widespread on the territory of Russia, Kazakhstan, and European countries. Praziquantel is the drug of choice for the treatment of opisthorchiasis, but this drug effects on O. felineus are poorly studied.
